  • GrowDex concentrations 0.5%, 0.8% and 1%. 
  • 384-well microtiter plates.   
  • 30 μl of GrowDex was dispensed per well and 30 μl of medium added on top of the gel. 
  • Aspiration and dispensing speed set at 10 μl/s. Blowout volume was set to 0 μl. 
  • 2000ms and 5000ms delays after aspiration and dispensing were tested. 
    • 2000ms better for GrowDex. 
  • Transfers done with Beckman Coulter Biomek AP384 P30XL tips (cat no. 22288).

Echo 525  LabCyte  Acoustic dispense (contact free transfer) 
  • GrowDex concentrations 0.2%, 0.4% and 0.6%.  
  • 384‐well microtiter plates. 
  • Transfer volume was 1.5 μl to 56 destination wells at 384­‐well plate.  
  • Echo is designed for the transfer of small volumes (25nl) droplets.  
  • GrowDex transfer fast and accurate – applicable for sequencing or qPCR purposes.

  • GrowDex concentration 0.4%. 
  • 96- and 384-well microtiter plates. 
  • Transferred volumes with standard 8-channel dispensing cassette.  
    • 50 μl per well for 96-wells. 
    • 25 μl per well for 384-wells.

  • GrowDex concentration 0.4%.  
  • 96- and 384-well microtiter plates. 
  • Transferred volumes with standard 8-channel dispensing cassette.  
    • 50 μl for 96-wells. 
    • 25 μl for 384-wells. 
  • 50 μl to 300 μl epTips® pipetting tips were used.

  • HepG2 cells dispensed into 96-well plates. 
  • GrowDex concentrations 0.7% and 0.8%. 
  • Biomek AP96 P250 tips (Beckman Coulter, USA). 
  • 100 μl of GrowDex was dispensed in each well and 100 μl of medium on top. 
  • Alamar blue and Live/Dead assays used.

  • Cancer cells dispensed into 96- and 384-well plates.  
  • GrowDex concentrations 0.2% and 0.4%. 
  • Speed of aspirating and dispensing was 8 μl/s. Blowout volume was set to 0 μl. 
  • Delay of 2000ms was set.  
  
Certus Flex  Fritz Gyger AG   Gyger micro valve technology and air pressure control (contact free transfer) 
  • Caki-2 cells were dispensed into 384-well plate (Cat no. 781091, Greiner).  
  • GrowDex concentrations 0.2%, 0.4% and 0.8% tested. 
  • The viability of the cells was measured 
  • Angled head allows dispensing liquids on the side of the wall rather than directly to well.

CyBiWell 384  Cybio  Pipette-based using disposable tips 
  • HepG2 cells in 30 μl GrowDex in 384-well plate. 
  • GrowDex concentration 0.5%.  
  • 50 μl of culture medium added on top of GrowDex.  
  • Cell seeding density 250 cells/μl 
  • 1600 compound library at 20μM compound concentration screened.
